tuberculosis (tb) is a growing unsolved health concern, as it is the main infectious cause of death in the globe. exposure to mycobacterium tuberculosis leads to active disease in only ~10% people. an effective immune response in remaining individuals stops m. tuberculosis multiplication. however, the pathogen is completely eradicated in ~10% people while others only succeed in containment of infection as some bacilli escape killing and remain in non-replicating (dormant) state (latent tuberculosis infection) in old lesions. the dormant bacilli can resuscitate and cause active disease if a disruption of immune response occurs. the ability of the pathogen to develop into a dormant state is the main obstacle in overcoming the disease. resuscitation-promoting factors (rpf) are mycobacterial proteins secreted by viable cells of mycobacterium tuberculosis stimulating (re)growth of dormant, non-replicating m. tuberculosis cells in vitro. recently, mukamolova et al., have demonstrated that sputum samples from untreated tuberculosis patients reveal a bacterial subpopulation that can be cultured only by addition of rpf. this study aims to reduce the time of detecting tuberculosis by adding culture filtrate from wild type mycobacterium tuberculosis h37rv (containing rpf) to the liquid medium middle brooke 7h9 broth. the study also emphasis on the efficiency of recombinant rpf in potentially inducing the growth of smear positive culture negative sputum samples. out of 55 samples analyzed rpf induced growth in 27 samples where as in 7h9 only 19 samples were reported to be positive. there was induction on the same day in rpf and 7h9 for 17 samples, induction in rpf alone for 8 samples and early detection in rpf for 2samples when compared to that of 7h9. this clearly indicated the induction of dormant bacilli by rpf in addition to early growth of bacilli.
