ablipase is one of the important biocatalsyt in industrial application. this is due to their ability specifically hydrolyze the triglyceride, and the product of reaction can be useful to produce many useful daily products. the aim of this study to isolate the lipase producing bacteria and manipulated it for lipase production. in this study, the lipase producing bacteria were isolated from the lake and soil from different places. from these isolation, the strains were screened by using tributyrin agar to identify strains that can hydrolyze tributyrin which is as a substrate. the strains that produce the big clearing zone were further the study to identify by using both physical and sequencing of 16s rrna gene. then a lipase enzyme produced through 48 hours fermentation and lipase activity was determined. from the 16s rrna, the strains that produce high lipase activity are burkholderia cepacia and pseudomanas putida. burkolderia cepacia which is rod shaped and motile gram-negative bacteria were produced 0.4cm clear zone for 24 hours incubation and found the maximum lipase activity was 16.12 u/ml in liquid medium at 30 hours fermentation. the other strains which are pseudomonas putida also rod shaped and motile gram-negative bacteria . p. putida was produced 0.9 cm for 24 hours incubation. this bacteria exhibited maximum lipolytic activity, 3.7u/ml at 0 hour fermentation. based on the finding of present study, b. capecia and p. putida are the potential lipase producing bacteria and this finding can be useful in prospect to increase the production industrial enzyme.stract
