a method for preparing a composition according to claim 1 suitable for use as a blood substitute, comprising the steps of: converting a solution of purified hemoglobin to the carboxy form by saturating the hemoglobin with carbon monoxide; dialyzing the resulting carboxy-hemoglobin solution against a normotonic solution to concentrate the hemoglobin to approximately 10 g/dl; reacting the carboxy-hemoglobin with oxidized adenosine triphosphate (o-atp) to effect predominantly intramolecular crosslinking among the hemoglobin molecules; reacting the carboxy-hemoglobin with oxidized adenosine (o-adenosine) to effect predominantly intermolecular crosslinking between the hemoglobin molecules; quenching the o-adenosine crosslinking reaction by adding reduced glutathione to the solution; and converting the hemoglobin from the carboxy- to the oxy- form by flushing the solution with oxygen.
